754 Therapeutic TNF inhibitors exhibit differential levels of efficacy in accelerating chronic cutaneous wound healing in vivo

Adalimumab is the only FDA- and EMA-approved treatment for moderate to severe hidradenitis suppurativa (HS), where etanercept certolizumab-pegol have been shown be inefficacious, suggesting that mechanism of action (MOA) adalimumab distinct in HS may contribute improved wound healing. We demonstrated adalimumab, but neither nor certolizumab-pegol, induces a healing profile vitro by regulating macrophage differentiation matrix metalloproteinase expression which underlie difference efficacy between various anti-TNF agents. To examine compare therapeutic TNF inhibitors chronic cutaneous vivo, murine model was established breeding Lepr+/db mice with human-TNFα transgenic mice. Four 6-mm full thickness excisional wounds were created on back hTNF+/+Leprdb/db followed administration agents (adalimumab, infliximab, golimumab, or certolizumab-pegol) vehicle. Wound area measurements photographs taken every 2nd day up 4 weeks assess size reduction. The vehicle group exhibited impairments In contrast, infliximab accelerated healing, showing significant improvement starting from 7 lasting throughout rest process. Golimumab less effective, along insignificant hTNF transgenic-Db/Db conclusion, our data suggest exhibit differential levels accelerating vivo. Additional studies are being undertaken explore differentiate MOA other current animal as well patients.
